Spheroid size and cell concentration with Elplasia MPc 500

1. Materials

  • Cells:Rat Mesenchymal stem cells (MSCs)
  • Medium:DMEM/F-12(1/1) [+]L-Glutamine, [+]2.438 g/L Sodium Bicarbonate, 12.5% FBS, Antibiotics

2. Methods

1. Preparation of cells

Cell density are adjusted, according to the following formula.

  • * Reference: cell density and cells in one micro-hole

2. Cell seeding

  1. Add 4mL of cell suspension to each device on the bottom or along wall.
  2. The plate was carefully placed into water-jacket incubator under 5% CO2 and 95% air 37℃
  3. Cells form spheroid in 3days.

Note: We recommend that plate is gently tiled in a back-and-forth and side-to-side manner to disperse cells, if cells are inclined to one part.

3. Collection
The spheroids have to be collected before the diameter of spheroids grow over 400μm.

  1. Prepare another dish or 6-well plate for collection.
  2. Liquid for collection such as PBS, medium was added in the dish or the well.
  3. Dip the bottom of the device onto the liquid.

3. Result
